Popis The main challenge in breast cancer treatment is metastatic disease. For estimation of the risk of metastatic disease, traditional prognostic factors like tumor size, tumor grade or lymph node status are currently used. However, the predictive value of these factors is not sufficient. Therefore there is an urgent need for new more reliable molecular markers with higher sensitivity and selectivity. This project is focused on identification and functional studies on proteins involved in low-grade breast cancer metastasis. For the discovery phase we use a combination of comparative proteomics methods with validation by qRT-PCR profiling at transcript level. On the basis of our preliminary results transgelin seems to be an interesting target for further functional studies. Altered expression of transgelin has been previously reported in various types of cancers, sometimes with controversial outputs and interpretation. Here we focus on its role in breast cancer metastasis. Our preliminary data suggest that transgelin should positively influence metastasis of the tumors by increasing the migratory properties of cells through its stabilizing role on their actin cytoskeleton.
