Unraveling a FNR-based regulatory circuit in bacterium Paracoccus denitrificans using proteomics, transcriptomic and bioinformatic approaches

Popis | The switch from aerobic to anaerobic respiration in the bacterium Paracoccus denitrificans is orchestrated by the action of three Fnr-type transcription regulators FnrP, NNR and NarR, which are sensors for oxygen, nitric oxide and nitrite, respectively. In this study, we aimed to discover the role of Fnr-family transcription regulators in the aerobic-semiaerobic switch at global level. Methods and Materials: We analyzed the protein composition of four Paracoccus denitrificans strains (wild type, FnrP-, NNRand NarR- mutant strains) grown aerobically, semiaerobically and semiearobically in the presence of nitrate using two-dimensional gel electropohoresis with MS/MS protein identification, with data validation at the transcript (qRT-PCR) and genome (FNR-binding sequences) levels. Results: Expression profiles were acquired using two-dimensional gel electrophoresis for 737 protein spots, in which 640 proteins gene products were identified using mass spectrometry. |
