Popis Homologous recombination (HR) is a process widespread in nature and essential to maintain the integrity of the genome via repair of DNA double-strand breaks (DSBs). Several models were proposed to understand the mechanism of double-strand break repair by HR, identify separate steps and role of the individual proteins involved. The Mus81/MMS4 complex is a DNA structure-specific endonuclease involved in replication fork stability and DNA repair. It seems from genetic experiments that Mus81/MMS4 could interact with Srs2, a helicase which on one hand prevents untimely recombination and on the other hand is necessary for certain pathways of homologous recombination. In our study, we confirmed the interaction between Mus81/MMS4 and Srs2. We then analyzed the stimulation of Mus81/MMS4 nuclease activity by Srs2 and we found out that this stimulation depends on the DNA subtrate involved in the reaction. Our experiments also indicate that the interaction between Mus81/MMS4 and Srs2 is specific
