Comparative Study of Complexation and Dissociation Kinetics of Yttrium(III) with DOTA-like Macrocylic Ligands

Popis | Bifunctional chelating ligands (BFC) sequestering radiometal ions are used in radioimunotherapy and diagnosis. BFC contain metal coordination unit (e.g. macrocycle) attached to biologically active molecules, e.g. monoclonal antibodies, small peptides, hormones, sugars, etc. The complexes should exhibit high thermodynamic stability and, of course, kinetic inertness under physiological conditions. Both properties are strongly influenced by degree of derivatization of macrocyclic skeleton and type of linker attached to biomolecule. In this contribution, the influence of structural variation of linker of BFC on kinetic properties was studied. Under experimental conditions (t = 25 oC, I = 0.1 M KCl, pH = 5.6-7.1, cY = 7*10-7 M, cL = 1.6*10-5 M), which are closed to conditions of 90Y radiolabelling in nuclear medicine, the empirical order in reactivity in respect to yttrium(III) was obtained: DO3AP > DO3APPrA > DOTA > DO3A > DO3APABn The results of complexation kinetics were compared with data obtained for 90Y complexation by DO3APABn followed by radiometric method (cY = cL = 1.2*10-3 M, t = 25 oC, pH = 5.54) and both are in a good agreement. The results of dissociation kinetics of yttrium macrocyclic complexes are comparable with other lanthanide complexes. |
