Popis The capillary electrophoresis (CE) is commonly use methods for separation and determination of different types of analytes. Another less common CE application is the study of interactions. CE based affinity interaction studies take advantages of native state of interacting partners without immobilization or labelling. The typical injection volumes in some CE analyses for the affinity studies are more than 10%. In several special CE applications, such as enzymatic or derivatization reactions, the large volumes of sample and reagents are mixed directly in the capillary. So, capillary serves as mixing and/or reacting cell and separation column at the same time. This leads to the even smaller sample consumption besides full automation of the assays. Developing an effective way of mixing reactants inside the limited space of a capillary is a challenging task preventing the wider practical implementation of this otherwise attractive concept. The main objective of this study was to test the TDLFP methodological approach of sample preparation for CE affinity application.
