Analysis of amino acids in various biological samples by means of CE-MS

Popis | Amino acids (AAs) and their metabolites can be used as objectives for targeted metabolomic analyses. The subject of this contribution is application of capillary electrophoresis coupled with mass spectrometry (CE-MS) for AAs analyses in various biological matrices as a spent culture medium collected after cultivation of embryos after in vitro fertilization, and human plasma and urine of gestational diabetes patients. Sample pretreatment, CE-MS method optimization and validation will be discussed, e.g. preconcentration techniques (pH mediated stacking and acetonitrile stacking), background electrolyte and organic modifier selection, nebulization gas pressure, and sheath liquid composition. Conditions were chosen with regard to the separation time and efficiency, the signal intensities and the peak areas. Analyses were performed with CE system with the bare fused silica capillary. Analytes were separated in 2 M formic acid without organic modifier under voltage applied in normal polarity. Samples were spiked with the internal standard methionine sulfone. MS detection was performed with qTOF instrument in the positive ionization mode. The CE and MS was connected via the sheath liquid co-axial ESI interface. Presented method is suitable for the metabolomic approach in various types of sample. |
