Description | The work evidenced that STAT3-induced over-expression of MDR1 is dependent on area assigned *824, so we can conclude that in this area is localized the crucial STAT3 binding site. Area *824 contains direct repeat with a three-nucleotide spacer and a three direct repeats with four-nucleotide spacer. Previous studies have shown that pregnane X receptor and constitutive androstane receptor, members of the nuclear receptor superfamily, as well as thyroid hormone receptors, mediate the up-regulation of MDR1 through this response region located -7.9 to -7.8 kb upstream of the MDR1 using an epithelial cell line LS174T or Caco2 (Burk et al., 2005; Geick et al., 2001; Kurose et al. 2008). This illustrates the general importance of this enhancer region upstream of MDR1 gene. |
