Description | Methicilin-resistant Staphylococcus aureus (MRSA) is a major nosocomial pathogen responsible for various infections both in health care and community settings. In this study we focused on detailed genotyping of 28 oxacilin-resistant strains isolated in Masaryk’s hospital in Ústí nad Labem in the period of increased incidence of MRSA strains. Methods: PFGE fragments of DNA digested with SmaI restriction endonuclease, spa typing, PCR detection of the type of SCCmec cassete, prophage carriage, detection of selected genes coding the virulence factors and genes mediating resistance to antibiotics and antimicrobial agents, analysis of plasmids and sensitivity test to polyvalent phage 812 were performed. Results: We have found that all strains harbored the SCCmec type II cassette and repetitions of the spa type t003. Strains differed from each other in PFGE patterns, prophage content and genes coding various virulence factors and resistance genes localized on plasmids. All strains were found to be sensitive to cytolytic action of the polyvalent phage 812. Conclusions: Genotypic analyses revealed a collection of related strains originated from the clonal lineage CC5. Our results contribute to molecular epidemiology of the MRSA in the Czech Republic as well as in neighbouring regions and countries in Europe. |
