Description Genes rpoN (coding for sigma 54 factor) and fhlA (coding for putative transcriptional activator) were cloned into the arabinose-inducible expression vector pBAD/HisB. Q-PCR technique was used for the study of putative gene regulation mediated by RpoN and FhlA in the clones of the BAC library. sigma 54-dependent promoters were identified in the T. pallidum genome using in silico approach and the predicted transcripts were mapped by PCR technique. Two possible sigma 54-dependent promoters (in the three BAC clones used) were identified by the software Promscan. Promoters were predicted upstream of sigA (TP1012) and TpF1 genes and transcript starts were confirmed by PCR amplifications. The experimental findings of the hlyIII and TpF1 (both organized in one operon) and sigA were in agreement with in silico analysis. Proteins RpoN and FhlA are positive regulators of gene expression of several treponemal genes.
