Description Human pluripotent stem cells (hPSC), albeit human induced pluripotent stem cells (hiPSC) or human embryonic stem cells (hESC), are a powerful tool in research and also hold great potential for clinical applications. One of their basic properties is active telomerase, which leads to telomere length maintenance or even lengthening in low passages (Zeng Sicong et al., 2014). Short telomeres can lead to early senescence in cells differentiated from hPSC. Therefore we used Southern blot analysis of terminal restriction fragments (TRFs) to test if the telomere length changes during reprogramming and culture of hiPSC in low passages. Contrary to previously published work (Yehezkel Shiran et al., 2011), we did not find any trend within the tested hiPSC lines. We next tested the possible impact of culture conditions on telomere length. We compared the telomere length of hPSC lines cultured for 15 passages in feeder-dependent and feeder-free systems. We also tested the effect of hypoxic/normoxic culture conditions. We did not prove any influence of culture conditions on the telomere length.
