Description Long-acting dosage forms are gaining in importance in psychopharmacology and are more effective than conventional p.o. dosage forms.The main benefits of long-acting psychotropic drugs include improved adherence to treatment, bypassing the first-pass effect and problems associated with absorption from the gastrointestinal tract, and the absence of a risk of accidental or intentional overdose. The development of long-acting dosage forms is currently focused on the study of microparticles. Based on preformulation experiments, suitable formulation and process parameters were selected for the preparation of microparticles based on lactic acid and glycolic acid copolymer (PLGA) for parenteral administration. The drug content, morphology and dissolution test of the prepared microparticles were evaluated. 6 different drug release profiles were prepared by appropriate procedures and subsequently 2 profiles were selected for in vivo application (laboratory rats). The pharmacokinetic profile of the administered microparticles was determined on the basis of frequent sampling using a previously developed and validated HPLC method.The serum was also used to determine liver tests, lipid spectrum and selected adipokines. According to the biochemical tests, we found a minimal effect on the levels of ALT, AST. In the medicated group, we observed slight differences from the control group in the lipid spectrum and levels of selected adipokines.
