Description Pharmacokinetics and pharmacodynamics behaviour of drug is influenced by its binding on human serum albumin (HSA) that is the most abundant protein in human blood plasma and it constitutes about half of serum proteins. Therefore the evaluation of analytical methods for analysis of these interactions with a small amount of sample solution is a highly desirable. In this work, four methodologies differing in their basis – capillary electrophoresis - frontal analysis (CE FA), equilibrium dialysis (ED), circular dichroism (CD) and isothermal titration calorimetry (ITC) were used for the determination of binding constant (Kb) as a criterion of the interaction strength. The main objective of this study was to determine Kb and to compare these methodological approaches. For this purpose, diclofenac HSA and lidocaine HSA were chosen as model systems. Obtained values of Kb values are comparable with values from literature.
