Elevated FGFR3 expression as a result of translocation t(4;14) in patients with multiple myeloma

Description | Translocation t(4;14) is adverse cytogenetic prognostic factor in patients with MM. Increased expression of FGFR3 gene as a result of t(4;14) (IgH enhancer juxtaposition) was also reported. We analysed FGFR3 gene expression in patients with/without t(4;14). Translocation t(4;14) was evaluated by FISH in MACS separated CD138+ cells obtained from 32 MM patients. Total RNA was transcribed into cDNA (Ambion WT Sense Target assay), labeled and hybridized to the Affymetrix GeneChip Human Gene ST 1.0 array. We anticipate, that the IgH enhancer might be deleted during ilegitimate switch recombination, thus t(4;14) did not always lead into FGFR3 overexpression and patients [with t(4;14)and without FGFR3 overexpression] might represent group of patients with favourable outcome. Our finding should be confirmed by survival analysis in larger cohort. |
