Description Genes rpoN (coding for 54 factor) and fhlA (coding for putative transcriptional activator) were cloned into the arabinose-inducible expression vector pBAD/HisB. Q-PCR technique was used for the study of putative gene regulation mediated by RpoN and FhlA in the clones of the BAC library. 54-dependent promoters were identified in the T. pallidum genome using in silico approach and the predicted transcripts were mapped by PCR technique. Proteins RpoN and FhlA are positive regulators of gene expression of several treponemal genes.
